With Locale, you create situations specifying conditions under which your phone's settings should change. For example, your "At School" situation notices when your Location condition is "77 Massachusetts Ave.," and changes your Volume setting to vibrate.

If you want to pin point your location you'd need GPS, otherwise it should get fairly close using the cell towers. Wifi shouldn't matter, it just needs to get out to the Internet, it can do that over the 3G connection. It also uses wi-fi positioning to get a better location, but I don't use the wi-fi part becasue I don't to have my wi-fi on all the time.
With GPS only, Locale did not activate the settings that I had set for each location. Used cell towers instead of the GPS and it worked great. The only issue is that cell tower location would be less accurate in that it would put you in a mode for a broader area than just the specific area that would be used with the GPS, if that makes sense. For example, if you use cell towers and want different settings at house and at church, which is close to your house then it may not be able to accomplish that because they both are in the range of the same cell tower.
